雄マウスから卵子、子も誕生 世界初、iPS細胞で―大阪大など

 雄マウスの人工多能性幹細胞(iPS細胞)から卵子を作り、子どもを誕生させることに、大阪大の林克彦教授らのチームが成功した。哺乳類の雄の細胞から卵子を作製したのは世界初という。研究成果の概要は25日までに英科学誌ネイチャーに掲載された。
 マウスや人の性別はXとYの性染色体の組み合わせで決まり、「XY」なら雄、「XX」なら雌になる。林教授らは、加齢によってY染色体が消え、X1本だけの細胞があることに着目。雄マウスから作製したiPS細胞を数代にわたって培養し、Y染色体のない細胞を作った上で、特殊な化合物を使って「XX」の細胞を作り出した。
 これを卵子に育てて別の雄の精子と体外受精させ、計630個を雌の子宮に移植。7匹の子どもが誕生し、全ての子が成長することも確認できた。
 この技術を活用するとX染色体を増やすことが可能となり、林教授は「(女性のX染色体が欠けて発症する)ターナー症候群などの治療法開発に役立つのではないか」と説明した。
 ただ、人の誕生に応用するには課題が多い。iPS細胞から人の卵子を作るには技術的に10年程度かかると見込まれるだけでなく、倫理的な議論が欠かせない。林教授は「技術の安全性を調べるのはわれわれの責任だが、どう使うかは社会に決めてもらうしかない」と話している。(2023/03/25-13:34)

Japan Team Creates Offspring Using Eggs from Male Mice Cells


A team of Japanese researchers has succeeded in creating mouse offspring from eggs made using induced pluripotent stem, or iPS, cells created from male mice.
   It is the first time in the world that eggs were produced from cells taken from male mammals. The research by the team, led by Osaka University professor Katsuhiko Hayashi, was published in British science journal Nature.
   The sexes of mice and humans are determined by the combination of X and Y chromosomes, with "XY" being male and "XX" female.
   Hayashi's team focused on the fact that the Y chromosome in male cells disappears with age, leaving the cells with a single X chromosome. The team cultured iPS cells created from male mice cells over several generations to produce cells without the Y chromosome, and then made them into cells with two X chromosomes using a special compound.
   The XX cells were grown into egg cells and fertilized in vitro with sperm from another male mice. A total of 630 such fertilized cells were then implanted in the uterus of female mice, resulting in the births of seven mice, all of which were later confirmed to have grown up.
